Abstract:
To further advance the capabilities of cryoEM, the CRYO ARM platform is undergoing a series of strategic updates aimed at improving performance, usability, and accessibility. Key hardware enhancements include the integration of a high-precision condenser lens (CL) aperture and a newly designed Koehler illumination system, both contributing to improved beam stability and image quality. These upgrades, combined with a new vacuum system utilizing roots pumps, significantly reduce maintenance frequency and operational costs while enhancing overall system durability.
To streamline data acquisition, the implementation of SmartScope - an AI-driven software developed at Princeton University - enables automated detection and selection of optimal squares and holes, thereby accelerating and simplifying the single particle analysis (SPA) workflow.
In addition, the adoption of high-resolution polepieces (HRP) has led to notable improvements in SPA performance, achieving sub-1.5 Å resolution. Complementary developments in cryoET and CryoLamellar workflows further expand the versatility of the CRYO ARM system, positioning it as a comprehensive solution for cutting-edge structural biology research.
